摘要:[目的]研究小麦成熟胚离体培养时的消毒处理。[方法]采用复合、单独使用消毒剂的消毒方法,对6个冬小麦品种进行消毒。[结果]高浓度升汞对种胚造成伤害,品种间有遗传差异。[结论]复合使用消毒剂处理的愈伤组织质量明显好于单独使用的,一定浓度的次氯酸钠对种胚愈伤组织诱导有促进作用;各个品种最佳处理不同,75%酒精1min+12%次氯酸钠8min+0.10%升汞12~18min效果比较好;利福平、多菌灵处理对小麦的种胚消毒起到一定作用,但出愈率有所下降。
[Objective] The research aimed to study sterilization treatments of Triticum aetivum mature embryo culture in vitro.[Method] The experiments were carried out by using compound and single disinfectants to six varieties of Triticum aetivum.[Result] High concentrations of mercuric chloride made damage to embryo,and there were genetic differences between breeds.[Conclusion] Quality of callus treated by using compound disinfectants showed better than single disinfectant obviously.A certain concentration of sodium hypochlorite could promote embryo callus induction.The best treatments to varieties were different.75% alcohol 1 min +12%NaClO 8 min +0.1% mercuric chloride 12 to 18 minutes was the best.The treatment by rifampicin,carbendazim played a certain role on disinfection of Triticum aetivum embryo,but callus formation declined.
