摘要:以抗病南瓜种质482-2的种子为材料,通过组织培养,研究并建立了离体再生体系,结果表明:南瓜种子无菌萌发后,分别以子叶、下胚轴和胚根为外植体进行诱导培养,子叶可直接分化出不定芽,下胚轴和胚根可产生愈伤组织、但很难进一步分化出不定芽,子叶分化出的不定芽经生根培养能获得完整植株;南瓜种子无菌萌发的适宜培养基为琼脂胨,子叶诱导不定芽的适宜培养基为MS+6-BA1.0 mg/L,不定芽生根的适宜培养基为MS+IBA0.2 mg/L。
Taking the seeds of disease resistant pumpkin germplasm 482-2 as material, the pumpkin regeneration in vitro was studied. The results showed that cotyledon could regenerate adventitious bud directly, but hypocotyl and radicle could be induced callus only, which was difficult to regenerate adventitious bud. After the rooting culture of adventitious bud, complete plantlet could be got. The research demonstrated that the suitable medium for pumpkin seed germination was agar medium, the suitable medium for cotyledon regenerating adventitious bud was MS+BA 1.0 mg/L, the suitable medium for buds rooting was MS+IBA 0.2 mg/L.
