摘要:山羊痘病毒属(Capripoxvirus,CaPV)成员包括牛结节性皮肤病病毒(Lumpy skin disease virus,LSDV)、山羊痘病毒(Goatpox virus,GTPV)和绵羊痘病毒(Sheeppox virus,SPPV),这三种病毒可跨界传播,已在全球大多数国家出现暴发或流行,严重影响了牛羊养殖业发展。由于CaPV成员的核苷酸相似性高达96%~97%,且不具有严格宿主特异性,一般检测方法很难鉴别CaPV成员,因此建立早期、快速、准确鉴别CaPV成员的方法将有助于阻止疫病传播。笔者综述了国内外CaPV成员检测方法的最新研究进展,包括病毒分离鉴定、病毒中和试验(virus neutralization test,VNT)、间接荧光抗体试验(indirect fluorescent antibody test,IFAT)、酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)和免疫过氧化物酶单层分析(immunoperoxidase monolayer analysis,IPMA)、聚合酶链式反应(polymerase chain reaction,PCR)、实时荧光定量PCR(real-time fluorescence quantitative polymerase chain reaction,qPCR)、环介导等温扩增试验(loop mediated isothermal amplification test,LAMP)、高分辨率熔解曲线分析(high resolution melting curve analysis,HRM)和重组酶聚合酶扩增(recombinant polymerase amplification,RPA)等,并论述不同检测方法的优缺点,以期为进一步探索CaPV成员检测方法提供参考。
Capripoxvirus(CaPV)includes lumpy skin disease virus,goatpox virus and sheeppox virus,which can spread across borders.It has broke out or spread in most countries around the world,which has seriously affected the economic development of cattle and sheep breeding industry.Because the homology of these three viruses is as high as 96%-97%,and they do not have strict host specificity,it is difficult to distinguish CaPV members by common detection methods in terms of clinical symptoms,viral structure and antigenicity.Therefore,the establishment of an early,rapid and accurate method for identifying CaPV members will help to prevent and control the spread of the disease.The author reviewed the latest research progress in the detection of CaPV members at home and abroad,including virus isolation and identification,virus neutralization test(VNT),indirect fluorescent antibody test(IFAT),enzyme-linked immunosorbent assay(ELISA),immunoperoxidase monolayer analysis(IPMA),polymerase chain reaction(PCR),real-time fluorescence quantitative polymerase chain reaction(qPCR),loop mediated isothermal amplification test(LAMP),high resolution melting curve analysis(HRM),and recombinant polymerase amplification(RPA).The advantages and disadvantages of different detection methods were discussed in order to provide reference for further exploration of CaPV members detection methods.
