摘要:为研究中甸犏牛溶质载体转运蛋白基因(SLC11A1)的结构变异情况,并评估结构变异与抗病性的关联,采集了114个样品,扩增了SLC11A1基因3'非翻译区(3'UTR)部分序列,测序共检测到3个微卫星多态位点(MS1、MS2、MS3)和3个核苷酸变异(1 782位点T→G、1 805位点G→A、1 907位点G→T),其中MS1有6个等位基因(GT10、GT12、GT14—17)、MS2有3个等位基因(GT5—7)、MS3有5个等位基因(GT12—16)。GT12/15、GT5/5、GT13/13基因型频率最高,分别为0. 204、0. 05、0. 324; GT15、GT5、GT13等位基因频率最高分别为0. 284、0. 685、0. 433。对MS1、MS2、MS3这3个多态位点进行单倍型分析,发现了44种单倍型,其中GT12/5/13为优势单倍型,频率为0. 13。χ2比较分析表明,中甸犏牛与婆罗门瘤牛×布兰科牛、荷斯坦牛×布兰科牛、布兰科牛、非洲牛MS1位点的GT12/12基因型频率差异显著(P <0. 01),与西班牙黄牛、荷兰黄牛MS3的GT13/13基因型频率差异显著(P <0. 01),与非洲黄牛差异不显著(P> 0. 05)。综上,中甸犏牛SLC11A1基因3'UTR具有丰富的多态性,推断其GT12/12和GT13/13纯合子基因型与抗病性相关。
In order to study the structural variation and theassociation with resistance to disease of the solute carrier transporter gene(SLC11A1)in Zhongdian yakow,114 samples were collected,and the 3′untranslated region(3′UTR)276 bp sequence of the SLC11A1 gene was amplified,and 3 microsatellite polymorphic loci(MS1,MS2,MS3)and 3 nucleotide variations(1 782 loci T to G,1 805 loci G to A and 1 907 loci G to T)were detected by direct sequencing.6 alleles(GT10,GT12,GT14-17)were detected at MS1,3 alleles(GT5-7)at MS2,and 5 alleles(GT12-16)at MS3.The frequencies of G12/15,G5/5 and G13/13 genotypes were the highest,0.204,0.05 and 0.324,respectively.The highest frequencies of GT15,GT5 and GT13 alleles were 0.284,0.685 and 0.433,respectively.Haplotype analysis of 3 polymorphic loci of MS1,MS2 and MS3 revealed 44 haplotypes,of which GT12/5/13 was the dominant haplotype with a frequency of 0.13.The chi square comparative analysis showed that the frequency of GT12/12 genotypes of Zhongdian yakow and Brahman Zebu×Blanco Orejinegro,Holstein×Blanco Orejinegro,Blanco Orejinegro,African cattle in MS1 was significantly different(P<0.01),and the GT13/13 genotype frequencies of Zhongdian yakow and Spanish cattle and Dutch cattle in MS3 were significantly different(P<0.01),and with African cattle,the difference was not significant(P>0.05).To sum up,this study revealed the rich polymorphism at 3′UTR of SLC11A1 gene in Zhongdian yakow.The preliminary assessment of the genotype of GT12/12 and GT13/13 homozygous genotypes in Zhongdian yakow was related to disease resistance.
