摘要:该文通过实验探讨了基本培养基及外植体的取材部位、PH、激素等因素对大扁杏离体组织茎芽分化诱导的影响,为完善大扁杏组培快繁体系提供了科学依据,从而为大量扩繁和优良品种的选配奠定了基础。结果表明:采用0.1%HgCl。对外植体消毒7min效果最佳;茎段初始培养选用改良MS(1/2NH4NO3)培养基最为理想;在改良的MS(1/2NH4NO3)培养基中同时添加1.0mg/L6-BA和0.08mg/LIBA时,茎芽增殖和伸长生长均达到最大;茎芽增殖和生长均达到最大的合适外植体为茎尖,PH为5.8时最佳。
The key techniques including explantation disinfection, basic medium, suitable explantation and PH are studied. By culturing the stems of Prunus Armeniacal Linn in vitro. The results showes as:using HgCl2 0.1% to disinfect for 7min can set the best method, germination rate reaches 79.2%. Improved MS(1/2NH4NO3)and MS medium are better than others for the beginning culturing of Prunus Armeniaca stems. It improved that when buds induces and grows, added different concentration, 6-BA and IBA in improved MS and improved MS mediums, will has different influence on propagation and growth . The concentration of 6-BA and IBA reach 1.0mg/L and 0.08mg/L is best for buds propagation and growth, and improved MS medium is better than MS medium. The optimal explantation are stems with double axillary buds and PH5.8.
