摘要:[目的] 探讨杜仲叶结构发育与绿原酸积累的关系。[方法] 应用荧光显微技术和HPLC化学分析方法研究杜仲叶在生长发育过程中组织结构和指标成分的变化规律。[结果]叶片发育早期,随着栅栏薄壁组织、海绵薄壁组织等叶肉细胞的分化发育,杜仲叶片中绿原酸含量不断增加。6月叶片组织结构发育成熟时叶片中绿原酸的含量达到最高,为1.153%。随着叶片的衰老,叶片中绿原酸的含量不断降低,至落叶前叶片中绿原酸的含量最低,为0.483%。在紫外光下,绿原酸储备在叶表皮、叶柄表皮和果实表皮周围,呈现淡蓝色荧光现象。[结论] 综合分析叶片生物量和绿原酸的含量,7-8月份采集杜仲叶较合适。
[Objective] The aim was to discuss the relationship between the structural development and chlorogenic acid accumulation of Eucommia ulmoides leaves. [ Method] The change laws of organization structure and index components of E. ulmoides leaves during growth and development process were studied by the method of fluorescence microscope technology and HPLC chemical analysis. [ Result] The chlorogenie acid content of E. ulmoides leaves was increased continuously with the differentiation and development of mesophyll cells such as palisade parenchyma and spongy parenchyma in the earlier of the leaf development. When the leaf organization structure was mature in June, the chlorogenic acid content in the leaves reached highest, being 1. 153% and that was decreased continuously with the leaf senescence. The chlorogenie acid content in the leaves reached lowest, being 0. 483% before deciduousness. The chlorogenic acid was stockpiled around the leaf epidermis and the petiole epidermis and the fruit epidermis and it presented the sky-blue fluorescence phenomenon under ultraviolet ray. [ Conclusion] Comprehensive analysis of the leaf biomass and chlorogenic acid content, July and August was suitable to pick E. ulmoides leaves.
