摘要:为了研究猪圆环病毒2型(Porcine circovirus type 2,PCV2)Cap蛋白能否模拟PCV2在体外刺激外周血单核细胞(peripheral blood mononuclear cells,PBMCs),使猪白细胞介素21(interleukin-21,IL-21)和IL-17A基因转录水平发生变化,试验根据IL-21和IL-17A的基因序列,设计引物,利用实时荧光定量PCR检测IL-21和IL-17A基因的转录水平。为进一步分析IL-21和IL-17A转录调节的机制,研究还通过实时荧光定量PCR检测了PD-L1的转录水平。结果表明:PCV2 Cap蛋白和PCV2在体外均能刺激PBMCs,导致IL-21、IL-17A和PD-L1基因的转录水平升高,其中PCV2 Cap蛋白导致的IL-21基因转录水平升高更多,几乎是PCV2刺激时表达量的2倍,IL-17A基因的转录水平基本和PCV2刺激时保持一致;PCV2 Cap蛋白能模拟PCV2刺激PD-L1基因转录水平发生变化,但是与PCV2感染时转录水平变化趋势并不完全一致,PCV2刺激时PD-L1的转录水平更高,预示着PD-L1基因与IL-21和IL-17A基因的转录可能存在一定的调节关系。
In order to determine the role of porcine circovirus type 2(PCV-2)Cap protein as a granular antigen,the transcription level changes of porcine interleukin-21(IL-21)and IL-17A in peripheral blood mononuclear cells(PBMCs)stimulated by PCV-2 and PCV-2 Cap protein in vitro were studied.According to the gene sequences of IL-21 and IL-17A,primers were designed and quantitative real-time PCR was used.To further study the regulation mechanism of IL-21 and IL-17A transcription,quantitative real-time PCR was also used to detect the transcription level of PD-L1.The results showed that both PCV-2 CAP protein and PCV-2 stimulated PBMCs in vitro,resulting in increased transcription levels of IL-21,IL-17A and PD-L1 genes;the transcription level of IL-21 gene induced by PCV-2 Cap protein increased more,almost twice the expression level stimulated by PCV-2,while the transcription level of IL-17A gene basically remained the same as that stimulated by PCV-2.PCV-2 Cap protein can simulate PCV-2 to stimulate changes in the transcription level of PD-L1 gene,butit is not completely consistent with the change trend during PCV-2 infection;when stimulated by PCV-2,the transcription level of PD-L1 is higher,which indicates that there may be a certain regulatory relationship between PD-L1 gene and the transcription of IL-21 and IL-17A genes.
