摘要:目的:优化竹黄多糖的发酵工艺。方法:培养竹黄菌菌丝体液,以菌丝干重和多糖得率为指标,分析竹黄多糖发酵过程中碳源、氮源、pH值对多糖积累的影响。结果:蔗糖、玉米粉、酵母膏均有利于菌丝的生长和多糖的形成。通过正交试验确定了竹黄多糖最佳发酵培养基:蔗糖3%、玉米粉3%、酵母膏0.5%、麸皮2%、(NH4)2SO40.2%、MgSO4.7H2O0.05%、KH2PO40.1%;在10L发酵罐上竹黄多糖含量最高达10g.L-1。结论:该工艺稳定,可为工业生产提供理论依据。
OBJECTIVE: To optimize the fermentation technology for polysaccharides in Shiraia barnbusicola . METHODS: Trough incubation of Shiraia barnbusicola mycelia, with dry weight of mycelia and yield of polysaccharides as index, the effeces of carbon source, nitrogen source, and pH on the accumulation of polysaccharides were analyzed. RESULTS: Saccharose, corn powder and yeast extract all contributed to the growth of mycelia and the formation of Polysaccharide, The orthogonal experiment results showed that the optimum fermention medium of polysaccharides in Shiraia barnbusicola were as follows: saccharose 3%, corn powder 3%, yeast extract 0.5%, bran 2%, (NH4) 2SO40.2%, MgSO4·7H2O 0.05%, KH2PO4 0.1%. And the amount of Shiraia bambusicola Polysaccharide can be extracted as high as 10g·L^-1 in the 10L fermentation tank. CONCLUSION: This optimized technology was stable in result, and it can provide theoretical basis for the industrial production of Shiraia bambusicola Polysaccharide.
