摘要:对来源于普通小麦的4种不同类型分子标记在黑麦的通用性进行比较,明确它们在黑麦扩增的有效性和多态性。335对引物对普通小麦品种兰考矮早八和黑麦进行PCR扩增,结果表明,有80对SSR、93对EST-SSR、33对EST-STS和98对PLUG引物可以在黑麦中扩增出清晰的DNA条带,分别占所用引物的80.0%、93.0%、94.3%和98.0%,表明基于小麦开发的这4种引物均可以用于黑麦遗传物质检测。进一步分析不同类型引物在普通小麦品种兰考矮早八和黑麦间的多态性,发现PLUG引物在二者间的多态比例最高,达到96.0%;SSR引物的多态比例相对较低,为86.0%;EST-SSR和EST-STS多态引物比例基本一致,分别为90.0%和91.4%。总的来看,来自小麦EST序列的引物(EST-SSR,EST-STS和PLUG)在黑麦的通用性和多态性均优于基因组SSR,其中PLUG引物在黑麦的通用性最好。
In this study, transferability of four kinds of molecular markers from bread wheat was compared in rye,which were to clear their effectives and polymorphism in the amplification of rye. The genomic DNA of a bread wheat cultivar Lankao Aizao 8 and a diploid rye were amplified using 335 pairs of primers. The results showed that 80 pairs of genomic-SSR primers, 93 pairs of EST-SSR primers, 33 pairs of EST-STS primers and 98 pairs of PLUG primers could amplify clear DNA bands in rye, which occupied 80.0%, 93.0%, 94.3% and 98.0% of four kinds of molecular markers, respectively. Such indicated that these four kinds of primers could be used to test chromatin of rye based on wheat development. Furthermore, we found that the polymorphism rates of PLUG and genomic-SSR markers were the highest(96.0%) and the lowest(86.0%) through analyzing the polymorphism of different types of primers between wheat cultivar Lankao Aizao 8 and rye, respectively, and nearly equal polymorphism rates(90.0% and 91.4%) were observed in EST-SSR and EST-STS markers. In general, the transferability of EST-SSR, EST-STS and PLUG markers which derived from wheat EST sequences was higher for rye than that of genomic-SSR. Besides, the utility of PLUG markers should be superior to other molecular markers used in this study.
