摘要:2010年初,已免疫过新城疫疫苗的新乡市某鸡场蛋鸡出现以消化道、呼吸道出血为主要症状的疑似新城疫病例。将病料经10日龄SPF鸡胚增殖分离病毒,命名为ND-XX10。提取其病毒基因组RNA,并以此为模板,RT-PCR扩增获得F基因主要功能区(1~631bp)片段并进行测序。结果显示,ND-XX10毒株F蛋白的裂解位点为112R-R-Q-K-R-F117,是典型强毒株氨基酸序列结构,第101位和121位氨基酸残基分别是K(赖氨酸)和V(缬氨酸),通过基因进化树分析,确定此分离毒株属于基因Ⅶe型。
To isolate field Newcastle disease virus(NDV) stains,10-day-old SPF chicken embryos was inoculated with suspected sample collected from disease chicken.The virus genomic RNA is extracted and the main region of F gene(1-631 bp) fragment was amplified by RT-PCR and identified by DNA sequencing.The results showed that a new isolate,namely ND-XX10,was obtained.The deduced amino acid sequence of the F protein of ND-XX10 isolate at the cleavage site was 112R-R-Q-K-R-F117,which is typically found in virulent NDV isolates.The amino acid residues at position 101 and 121 are K and V,respectively.Phylogenetic analysis showed that the isolate belonged to genotype Ⅶe.
