摘要:bZIP转录因子在植物应对病虫害、病原菌、干旱、低温等胁迫中发挥着重要功能,然而金银花中该家族基因的功能报道较少。为了研究bZIP转录因子在金银花发育及非生物胁迫中的功能,本研究以‘金丰1号’为试验材料,研究了bZIP基因LjbZIP1的序列和表达模式。结果表明,LjbZIP1基因的开放阅读框长1 236 bp,编码411个氨基酸,预测分子量约为45.82 kD,等电点为7.30。LjbZIP1含有一个保守的BRLZ结构域。亚细胞定位预测LjbZIP1蛋白定位于细胞核。保守结构域同源性比对和进化树分析表明,LjbZIP1和拟南芥At1G19490在一个分支。组织特异性表达模式分析显示LjbZIP1在所有检测组织中均有表达,表达量由高到低分别为叶片,根和花,暗示LjbZIP1基因可能参与金银花叶片发育;干旱和盐胁迫处理分析表明,该基因能够被干旱和盐胁迫诱导表达。推测LjbZIP1可能调控非生物胁迫耐性。本研究为更深入揭开此基因在金银花发育及处置来自非生物胁迫反应中的功能提供帮助。
bZIP transcription factors play important roles in plant under various biological and abiotic stresses.However,the function of this family gene in Lonicera japonica is less reported.In order to study the function of bZIP transcription factor in Lonicera japonica development and abiotic stress,the gene sequence and expression pattern of LjbZIP1 were studied in this study with‘Jinfeng1’as the experimental material and by quantitative RTPCR.The results showed that LjbZIP1 gene had an open reading frame length of 1236 bp,encoded 411 amino acids,predicted molecular weight of 45.82 kD and isoelectric point of 7.30.LjbZIP1 contains a conserved BRLZ domain.Subcellular localization predicted the localization of LjbZIP1 protein in the nucleus.By means of conserved domain homology comparison and evolutionary tree analysis,LjbZIP1 and Arabidopsis At1G19490 were proved to be in a branch.Analysis of tissue-specific expression pattern showed that LjbZIP1 was expressed in all detected tissues,with the highest expression in leaves,followed by roots and the lowest in flowers,suggesting that LjbZIP1 gene may be involved in the development of leaves.Combined with the results of evolutionary tree analysis,it is speculated that LjbZIP1 may regulate abiotic stress tolerance.Drought and salt stress analysis showed that LjbZIP1 could beup-induced by drought and salt stress.This study could be helpful for further analysis of the function of this gene in Lonicera japonica development and abiotic stress response.
