摘要:为了研究兔脑炎原虫性脑肉芽肿形成与PI3K/AKT信号通路的关系,对患脑炎原虫病兔和正常对照兔的脑组织进行了组织切片和特异性蛋白提取,并用普通染色、特殊染色、免疫组织化学技术和Western blot技术分别对脑炎原虫性脑肉芽肿病变进行了观察;对病兔和对照兔脑组织内与PI3K/AKT信号通路中血管内皮生长因子(VEGF)、磷脂酰肌醇3激酶的调节亚单位P85(P85)、催化亚单位P110(P110)、蛋白激酶B(AKT)和磷酸化蛋白激酶B(p-AKT)等关键性蛋白的表达丰度进行了检测。结果显示,病兔的脑组织中均有不同形态的脑肉芽肿形成和非化脓性脑炎变化;免疫组化染色显示,VEGF、P85、P110、AKT和p-AKT的表达在组成肉芽肿中的巨噬细胞、上皮样细胞、星形胶质细胞、血管内皮细胞和成纤维细胞中明显增高,呈强阳性反应;Western blot检测结果显示,病兔脑组织中的VEGF、P85、P110、AKT和p-AKT等信号通路关键蛋白表达丰度明显高于健康兔,差异极显著。结果表明,PI3K/AKT信号通路在兔脑炎原虫性脑肉芽肿形成的形成过程起到重要作用;正是由于PI3K/AKT信号通路的激活才有效地促进了脑肉芽肿中各种细胞的增殖。
In order to study the relationship between cerebral granulomatous formation by Encephalitozoon cuniculi(E.cuniculi)and PI3 K/AKT signal pathway,the common dyeing,special staining,immunohistochemistry and Western blot were used to investigate the morphological changes of cerebral granuloma caused by E.cuniculi,and analyze the expressive abundance of five kinds of key proteins,such as VEGF,P85,P110,AKT and p-AKT in PI3 K/AKT signal pathway in the sections and protein extraction of brain tissues of rabbits infected with E.cuniculi and control rabbits.The results showed that there were numbers of cerebral granuloma and nonsuppurative encephalitis in all kinds of sick rabbits.The expression of VEGF,P85,P110,AKT and p-AKT was obviously higher in macrophages,epithelioid cells,astrocytes,vascular endothelia and fibroblasts in the cerebral granuloma.These cells showed strong positive reaction and were stained as dark brown by immunohistochemistry staining.The key protein abundance of signal pathway in brain tissue of the sick rabbit was much higher than that of the control rabbit by Western blot.In conclusion,the PI3 K/AKT signal pathway takes part in the formation of cerebral granuloma by E.cuniculi,and the proliferation of all kinds of cells in cerebral granuloma is related to the activation of PI3 K/Akt signal pathway.
